I actually asked my doctor how to go about changing pill-time from morning to evening and he said he saw no problem with doing that over the space of a few days.

For all your questions, consider this: The half-life of Harvoni in the body (can't remember the exact source, but it's mentioned in www.rxlist.com/harvoni-drug/clinical-pharmacology.htm )

So after one full day the concentration in your body would be roughly 75%, after 12h about 87% (I say roughly because it's logarithmic, not linear, but that should be good enough). You can see that changing your time of ingestion by 30 min makes virtually no difference, and missing one pill not too much.

1) no, but I anticipated that I might want to change the time. Instructions were to spread it out, NOT overdose.

2) don't know yet, 4week post EOT coming up, but how could anybody tell you if those changes would have had an effect? You'd have to clone yourself and try it both ways to see if there'd be any difference!

for 3) and 4) : That's not how resistance develops. Don't worry about that.

5) see above.

Also, keep in mind that treatment length and dosage are 'best guesses', not exact science. If it were, Harvoni wouldn't be prescribed in a one-size-fits all daily dosage for all body weights.

I usually took my pill at 9.30am, however by mistake I overdosed at 8.30pm (because that's when I take my 2nd of Ribavirin, accidentaly took a Harvoni too) so then my time Harvoni time became 8.30pm.

Basically an accidental time change.

 

1) Did you change the time you took your Harvoni? How? Did you overdose or took the pill later?

2) Did you still SVR?

3) Do you think resistance builds up if you overdose?

4) Do you think resistance builds up if you miss your pill by 30 minutes?

5) How about if you intentionally miss your dose by 30 minutes each day for 24 consecutive days?

I'm thinking of changing my dose back to 9.30am, but to do that I'd have to increase the time (so as to not overdose again, and effectively lose another pill in the process) between pills. I'm thinking of doing that by intentionally taking the pill later by 30 minutes for 24 consecutive days (so in total that's 12 hours). This would imply me having to wake up in the middle of the night to take the pill for a while.
