I believe Sovaldi and Olysio, while a very effective treatment option, has been wrought with problems. It's biggest obstacle has been the fact that two different pharmaceutical companies own the patent rights to them. As is often the case in blockbuster medical treatments, the driving force turns from joint cooperation, to individual greed and such has been the path of these two medications. As Gilead gets ever closer to the FDA approval of Ledispavir, the attention to the off label administration of Sovaldi and Olysio will disappear because Sovaldi and Ledispavir are both manufactured by Gilead and reports are already showing higher rates of success than does Sovaldi and Olysio combined. Sov/Led will also as a result of the spanking Gilead has taken over the price rollout of Sovaldi alone, be released at a much lower price than the $148K retail price of Sov/Oly now. It's always about the money... no

After 4 weeks on the Solv/Olys combo, the virus is undectable and liver enzymes are almost normal range. At this point, nausea and headaches are my only side effects, with the expected fatigue. We are so fortunate to have these treatments available. Good luck to all!

G1a prior tx 2005 with peg & RIBA -quit due to bad side effects. Follow up visit showed it was back. Current tx Olysio/sovaldi 673,000 vl on 3/21/14. May 1st undetected vl. EOT 12 weeks undetected!

Great news!  I am mostly likely going to be in the Optimist trial which is either an 8 or 12 week.  I'm curious Mallini why you felt the Solvadi/Olysio combo will be a thing of the past?

These are good results, but remember they are from a Phase 2 Trial, with small numbers.

Janssen is funding the Phase 3 Optimist Trials with Sovaldi/ Olysio but no Ribavirin. The numbers total 400 patients, which is small for a Phase 3 Trial. My feeling is that this combo will be a thing of the past by the time the results are out.

Gilead will price Sovaldi/Ledipasvir below the Sovaldi/Olysio cost( which is $148K for 12 weeks). It's no contest.

Good Luck Gracie on getting started. This treatment is really side effect free for me ...feels like I'm not taking anything. You'll be pleasantly surprised...guaranteed! 

BigPharma, treatment is going great. Haven't had any VL tests yet.  I can guess what will happen.. VL UND.. I hope anyway. That seems to be the case with everyone on this treatment. I go for my tests on May 1st and will post my results.

Love love love to read things like this. Now to get my doctor on board. My nurse is soon going on maternity leave, and here in Canada that means she's off for a year with nobody so far to take her place. I may have to travel outside of my area to get a hepologist to treat me, which of course I will. 

....keep up the great work peeps.... The news is so positive on here wimock success after success ... Yahooooo!

Sovaldi/Olysio Combo Cures 94% of Those With Liver Disease

 

 

In a recent trial, combination therapy with Gilead Sciences Sovaldi (sofosbuvir) and Janssens Olysio (simeprevir) cured 94 percent of people who had genotype 1 of hepatitis C virus (HCV) and advanced liver disease, MedPage Today reports. Eagerly anticipated results from the Phase II COSMOS trial were presented at the 49th annual meeting of the European Association for the Study of the Liver (EASL) in London.

Eighty-seven study participants with F3 or F4 METAVIR fibrosis scores were divided into four groups: One took the NS5B nucleotide polymerase inhibitor Sovaldi plus the NS3/4A protease inhibitor Olysio for 12 weeks, and the other took the combination for 24 weeks; two other groups added ribavirin to the regimen and were treated for either 12 or 24 weeks.

Neither treatment length nor ribavirin use affected the success rate. In the 24-week groups, 93 percent (28/30) of those who took ribavirin achieved a sustained virologic response 12 weeks after completing therapy (SVR12, considered a cure), compared with 100 percent (16/16) who did not take ribavirin. In the 12-week groups, 93 percent (25/27) of those who took ribavirin were cured, compared with 93 percent (13/14) of those who did not take ribavirin.

Notably, those who had the Q80K polymorphism had the same outcomes as those who did not. Past research has found that the polymorphism lowers cure rates among those treated with protease inhibitors. Sovaldi apparently canceled out this effect.
